Anatomical Study of Somatic Embryogenesis in Glycine max (L.) Merrill BABT

A comparative anatomical analysis of somatic embryogenesis in two soybean (Glycine max (L.) Merrill) genotypes was carried out. The somatic embryos were originated from cotyledonary explants obtained from immature zygotic embryos. The medium used for somatic embryogenesis induction was Murashige and Skoog, 1962, salts and Gamborg et al., 1968, vitamins (MSB) supplemented with 0.8 mg.L-1 of 2,4-D for genotype PI 123439 and 40 mg.L-1 of 2,4-D for ‘Williams 82’. Cytogenetic studies in some species of Passiflora L. (Passifloraceae): a review emphasizing Brazilian species BABT

The Passifloraceae is represented by species of tropical and subtropical origin. The Passiflora is the richest genus with approximately 450 species, 200 of them being native to Brazil. Recent karyological studies have reported the basic chromosome number for the Passiflora genus as x = 6, whereas x = 9, x = 10 and x = 12 were established as secondary basic numbers. High rates of fertility occur in most Passiflora species, since both meiotic index and pollen viability are above 90%. Unusual meiotic behavior has been described in some taxa. Genetic instability of sugarcane plants derived from meristem cultures Genet. Mol. Biol.

The RADP (Random amplified polymorphic DNA) technique was used to detect tissue-culture-induced variations in sugarcane. Plants of the Brazilian variety RB83-5486 propagated via rhizomes and via meristem cultures were studied. The polymorphism rate for 98 RAPD loci was 6.93% when the plants derived from meristems. Besides, in order to evaluate the influence of the number of subcultures on the generation of somaclonal variation, field-grown RB83-5486 plants derived from 10 meristems were studied after five subcultivations. Although different rates of polymorphism were observed, there was no direct association with the stage of subcultivation. Genetic variability in the endophytic fungus Guignardia citricarpa isolated from citrus plants Genet. Mol. Biol.

During some phases of of their life-cycle endophytic fungi colonize plants asymptomatically being found most frequently inside the aerial part of plant tissues. After surface disinfection of apparently healthy leaves from three varieties of mandarin orange and one tangor, and after incubation on appropriate culture medium, 407 fungal isolates were obtained, giving a total infection frequency of 81%. No fungal growth was observed from disinfected seeds, indicating that fungi are probably not transmitted via seeds. Isoenzymatic variability in tropical maize populations under reciprocal recurrent selection Scientia Agricola

Maize (Zea mays L.) is one of the crops in which the genetic variability has been extensively studied at isoenzymatic loci. The genetic variability of the maize populations BR-105 and BR-106, and the synthetics IG-3 and IG-4, obtained after one cycle of a high-intensity reciprocal recurrent selection (RRS), was investigated at seven isoenzymatic loci. A total of twenty alleles were identified, and most of the private alleles were found in the BR-106 population. One cycle of reciprocal recurrent selection (RRS) caused reductions of 12% in the number of alleles in both populations. In order to verify whether genetic distance (GD) is associated with population mean (PM), genetic variance (GV) and the proportion of superior progenies generated by each cross in advanced generations of selfing (PS), the genetic distances between eight soybean lines (five adapted and three non-adapted) were estimated using 213 polymorphic RAPD markers. The genetic distances were partitioned according to Griffing's Model I Method 4 for diallel analysis, i.e., GDij = GD+ GGDi+ GGDj + SGDij. Screening of Passiflora species for reaction to Cowpea aphid-borne mosaic virus reveals an immune wild species Scientia Agricola

Cowpea aphid-borne mosaic virus (CABMV) is a potyvirus that causes the most serious virus disease of passion fruit crops in Brazil. It is transmitted by several species of aphids in a non-persistent, non-circulative manner. The reaction of 16 species of Passiflora to infection by mechanical inoculation with four Brazilian isolates of CABMV was evaluated under greenhouse conditions. Only P. suberosa, a wild species, was resistant to infection by all virus isolates, in two independent assays. P. suberosa grafted onto infected P. edulis f. flavicarpa did not develop symptoms; neither was the virus detected by RT-PCR in the upper leaves, suggesting that this species is immune to CABMV.

Somatic hybridization between Citrus sinensis (L.) Osbeck and C. grandis (L.) Osbeck PAB

This work had as objective to produce citrus somatic hybrids between sweet oranges and pummelos. After chemical fusion of sweet orange embryogenic protoplasts with pummelo mesophyll-derived protoplasts, plants were regenerated by somatic embryogenesis and acclimatized in a greenhouse. The hybrids of 'Hamlin' sweet orange + 'Indian Red' pummelo and 'Hamlin' sweet orange + 'Singapura' pummelo were confirmed by leaf morphology, chromosome counting and molecular analysis. These hybrids have potential to be used directly as rootstocks aiming blight, citrus tristeza virus, and Phytophthora-induced disease tolerance, as well as for rootstocks improvement programs.

Foram conduzidos experimentos visando à otimização de procedimentos para preparações citológicas de pontas de raiz com alta freqüência de metáfases mitóticas, apresentando cromossomos com morfologia nítida, em espécies pertencentes a dois gêneros de plantas, Passiflora (Passifloraceae) e Crotalaria (Leguminosae). Para o acúmulo de metáfases, bem como de prófases e prometáfases com cromossomos bem condensados, foram avaliados pré-tratamentos de raízes, em que foram utilizadas diferentes combinações de 8-hidroxiquinolina, inibidor do fuso mitótico, com cicloheximida, inibidor da síntese protéica. Genetic transformation though protoplast electroporation has been established for commercially important plant species. In this work, explant sources, electric field strengths, electroporation buffers, DNA forms and osmotic pretreatment were assayed in order to optimize transient reporter gene expression in electroporated tissues of Stylosanthes guianensis, a tropical forage legume. Higher transformation rates were obtained employing cotyledonary explants and an electric field strength of 250 V cm-1.
